Functional characterization of HIV-1 Tat exon-1 variants from North India and their implications on HIV-1 transactivation and TAR interaction

چکیده
Background HIV-1 virus is a rapidly evolving virus due to genetic variability through its rapid replication, mutation and recombination potential which is a major hurdle in vaccine development. One of the effective ways to modulate HIV-1 infection is to target viral proteins; among the viral proteins, Tat plays a major role in HIV-1 pathogenesis. It activates viral gene expression through TAR interaction. The aim of this study was to characterize genetic and functional variants of Tat exon-1 from HIV-1 patients from North India.

Molecular and Genetic Characterization of Natural HIV-1 Tat Exon-1 Variants from North India and Their Functional Implications
BACKGROUND Designing an ideal vaccine against HIV-1 has been difficult due to enormous genetic variability as a result of high replication rate and lack of proofreading activity of reverse transcriptase leading to emergence of genetic variants and recombinants. Tat transactivates HIV-1 LTR, resulting in a remarkable increase in viral gene expression, and plays a vital role in pathogenesis. The ...
متن کاملImpact of Genetic Variations in HIV-1 Tat on LTR-Mediated Transcription via TAR RNA Interaction
HIV-1 evades host defense through mutations and recombination events, generating numerous variants in an infected patient. These variants with an undiminished virulence can multiply rapidly in order to progress to AIDS. One of the targets to intervene in HIV-1 replication is the trans-activator of transcription (Tat), a major regulatory protein that transactivates the long terminal repeat promo...
متن کاملEffect on HIV-1 Gene Expression, Tat-Vpr Interaction and Cell Apoptosis by Natural Variants of HIV-1 Tat Exon 1 and Vpr from Northern India
BACKGROUND Since HIV-1 Tat and Vpr genes are involved in promoter transactivation, apoptosis, etc, we carried out studies to find out nature and extent of natural variation in the two genes from seropositive patients from Northern India and determined their functional implications. METHODS HIV-1 tat exon 1 and vpr were amplified from the genomic DNA isolated from the blood of HIV-1 infected i...
متن کاملAmino acid sequence divergence of Tat protein (exon1)of subtype B and C HIV-1 strains: Does it have implications for vaccine development?
Functional genes of HIV-1 like the tat express proteins essential for viral survival and propagation. There are variations reported in levels of Tat transactivation among the different subtypes of HIV-1. This study looked at the amino acid differences in the different regions of Tat protein (exon 1) of subtype B and C strains of HIV-1 and tried to observe a molecular basis for protein function....
متن کاملCharacterization of natural HIV-1 Tat and Vpr variants from Northern India
Methods HIV-1 Tat exon 1 and Vpr were amplified from the DNA isolated from blood of HIV infected patients and cloned. Clones were got sequenced and aligned against reference sequences using CLUSTAL W. Sim plot analysis was done for recombinants. Their expression was accessed by transfection of HEK 293T cells with myc fusion clones of variants and western blotting using anti-myc antibody. The va...
